IRAK-4 (interleukin 1 receptor-associated kinase 4) belongs to the IRAK family, a family of protein kinases involved in innate immune response signaling from Toll-like receptors. There is evidence that IRAK4 is involved in three components of TLR signaling. First, IRAK4 is the initial kinase in the vicinity of TLR receptors and is used to activate downstream effectors in the inflammatory cascade response, such as cytokines and chemokines. Second, deletion of the IRAK4 gene leads to defects in various cytokine responses, and finally, IRAK4-deficient patients exhibit immunodeficiency to IL-1, IL-8, and other TLR-binding ligands.
